George Medicines (搜索) announced positive results from the investigator-initiated TRIDENT trial at the World Stroke (搜索) Congress 2025 in Barcelona, demonstrating that GMRx2 significantly reduced the risk of recurrent stroke when added to standard of care compared to placebo. The study met its primary endpoint, showing a statistically significant and clinically meaningful reduction in recurrent stroke among patients with a history of intracerebral hemorrhage (搜索) (ICH).
Trial Design and Patient Population
TRIDENT was a multicenter, international, double-blind, placebo-controlled, parallel-group, randomized controlled trial conducted in patients with a history of acute stroke (搜索) due to intracerebral hemorrhage (搜索) on standard of care treatment. The primary outcome was the time to first occurrence of recurrent stroke. The trial was led by The George Institute for Global Health and funded by the National Health and Medical Research Council (NHMRC) Australia and the Brazilian Ministry of Health, with study medication provided by George Medicines (搜索).
Clinical Significance for High-Risk Population
Acute ICH accounts for at least 10 percent of the 20 million new strokes worldwide each year. ICH survivors face high risk of recurrent stroke (搜索) and other serious cardiovascular events. Despite numerous studies indicating benefits of blood pressure-lowering medications for stroke survivors, many patients either receive no blood pressure-lowering medication or achieve inadequate control.

GMRx2 Triple Combination Therapy
GMRx2 is a combination tablet containing telmisartan (an angiotensin II receptor (搜索) blocker), amlodipine (a dihydropyridine calcium channel blocker), and indapamide (a thiazide-like diuretic). The TRIDENT trial evaluated the 20 mg/2.5mg/1.25mg formulation given once daily in addition to standard of care. The drug is also available in 10/1.25/0.625 mg and 40/5/2.5 mg dosage forms for hypertension (搜索) treatment.
Safety Profile and Previous Clinical Evidence
The safety profile of GMRx2 in the TRIDENT trial was generally consistent with the well-established safety profile of the medicine and its individual components. The drug's development is supported by a comprehensive clinical program, including two pivotal Phase III studies in hypertension (搜索) published in 2024 in the Journal of the American College of Cardiology and The Lancet.
In these previous trials, the triple combination demonstrated significantly reduced blood pressure and improved blood pressure control rates compared to dual therapy and placebo. Both trials showed good tolerability with no increase in withdrawal from treatment due to adverse events. GMRx2 was also investigated in the Nigerian VERONICA trial, which reported better blood pressure lowering with good tolerability compared to standard of care protocol.
First-of-Kind Trial in ICH Population
TRIDENT represents the first stroke (搜索) outcomes trial with a low-dose single pill triple combination therapy for blood pressure treatment in adults with a history of ICH on top of standard of care. This addresses a significant gap in evidence-based treatment options for this high-risk patient population.
Full results from the trial will be submitted for publication in a peer-reviewed journal. The company plans to leverage these findings to advance the clinical development and potential regulatory submissions for GMRx2 in stroke (搜索) prevention applications.
