Kamada Discontinues Phase 3 Inhaled AAT Trial Following Futility Analysis, Maintains Strong Financial Outlook
核心洞察
Kamada Ltd. discontinued its Phase 3 InnovAATe trial for inhaled alpha-1 antitrypsin (搜索) (AAT) therapy after an independent Data and Safety Monitoring Board determined the treatment was unlikely to demonstrate statistically significant benefit in its primary endpoint of lung function improvement.
The discontinuation was based solely on efficacy concerns following a prespecified interim futility analysis, with no safety issues identified during the trial.
Despite the trial setback, Kamada reaffirmed its 2025 revenue guidance of $178-182 million and adjusted EBITDA of $40-44 million, while projecting double-digit growth in revenues and profitability for 2026.
Kamada Ltd., a global biopharmaceutical company specializing in plasma-derived therapies, announced the discontinuation of its Phase 3 InnovAATe clinical trial evaluating inhaled alpha-1 antitrypsin (搜索) (AAT) for treating Alpha-1 Antitrypsin Deficiency (搜索) (AATD (搜索)). The decision followed recommendations from an independent Data and Safety Monitoring Board (DSMB) based on a prespecified interim futility analysis indicating the treatment was unlikely to achieve statistically significant improvement in its primary endpoint of lung function measured by FEV1 (搜索).
Trial Discontinuation Details
The discontinuation was solely related to the low likelihood of demonstrating efficacy and was not driven by safety concerns. The futility analysis was conducted as part of the trial's predetermined monitoring plan, designed to assess whether continuing the study would be likely to yield positive results.
"While we are disappointed that the trial did not pass this milestone and is being discontinued, we are well positioned to continue and support our 2026 and future growth prospects," said Amir London, CEO of Kamada.
Continued Commitment to AATD Community
Despite the trial setback, Kamada emphasized its ongoing commitment to patients with alpha-1 antitrypsin deficiency (搜索) through its existing commercial product GLASSIA®, an intravenous AAT treatment. The company continues to supply GLASSIA® internationally, including in the United States and Canada through a licensing agreement with Takeda (搜索).
Strong Financial Performance and Outlook
Kamada reaffirmed its robust financial position and growth trajectory. The company reiterated its 2025 full-year revenue guidance of $178 million to $182 million and adjusted EBITDA guidance of $40 million to $44 million, based on consistent strong performance throughout 2025.
Looking ahead to 2026, Kamada projects double-digit growth in both revenues and profitability. This projected growth is supported by the company's robust commercial portfolio, which includes six FDA-approved specialty plasma-derived products marketed in over 30 countries, a growing biosimilar portfolio in Israel, and expansion of plasma collection capacity.
Commercial Portfolio and Growth Strategy
Kamada's commercial portfolio encompasses six FDA-approved specialty plasma-derived products: KEDRAB®, CYTOGAM®, GLASSIA®, WINRHO SDF®, VARIZIG®, and HEPAGAM B®, along with KAMRAB®, KAMRHO (搜索) (D)®, and two types of equine-based anti-snake venom products. The company also maintains a distribution segment portfolio, primarily focused on launching biosimilar products in Israel.
The company operates three plasma collection centers in the United States, located in Beaumont, Houston, and San Antonio, Texas. This plasma collection infrastructure supports revenue growth through sales of normal source plasma to other manufacturers and meets increasing demand for hyper-immune plasma.
Strategic Focus on Business Development
Moving forward, Kamada plans to leverage its strong cash position to pursue new commercial-stage business development opportunities. The company's strategy focuses on securing significant new business development, in-licensing, collaboration, and merger and acquisition opportunities to enhance its marketed products portfolio and drive long-term growth.
Management indicated that detailed 2026 guidance will be provided in January 2026, as the company continues to focus on its three primary growth pillars: organic growth from commercial activities, strategic business development opportunities, and expansion of plasma collection operations.
