NeuroTherapia (搜索), Inc., a clinical-stage biotechnology company spun out of the Cleveland Clinic, has successfully completed its Phase 2a clinical trial of NTRX-07, an oral small molecule therapy for Alzheimer's disease. The 28-day double-masked, randomized trial enrolled 48 patients across five sites in Hungary, Poland, and the Czech Republic, achieving its primary objectives of demonstrating safety, tolerability, and target engagement.
Trial Design and Primary Outcomes
The Phase 2a study was designed to assess target engagement through biomarkers of neuroinflammation and neuronal function, while monitoring safety, pharmacokinetics, and standard clinical efficacy measures including ADAS-cog, MMSE, and Trail Making Test. The trial utilized a 1:1 randomized design and was managed by CRU Global (搜索), a leading European contract research organization.
"We are pleased to see that NTRX-07 continues to be safe and well tolerated in patients with Alzheimer's Disease, and that drug exposure levels were in line with what we expected, demonstrating a positive outcome," commented Dr. Joseph Foss, the company's Chief Medical Officer.
The primary objective of safety and tolerability was achieved, with overall adverse events similar across treatment and placebo groups. Notably, no treatment-related serious adverse events were reported in the active treatment arm.
Biomarker and Imaging Results
Preliminary analysis of exploratory endpoints revealed encouraging results across multiple domains. MRI findings were directionally supportive, with changes trending toward stabilization on active treatment, supporting the hypothesis that neuroinflammation plays an important role in Alzheimer's disease progression.
Dr. Foss noted that "following the preliminary analysis of the exploratory endpoints, the study met its core Phase 2a objectives and produced a positive multi-domain signal package across safety, PK, biomarkers, MRI, EEG, and clinical outcomes."
NeuroTherapia (搜索) is continuing collaboration with Oxford Brain Diagnostics Ltd (搜索) for more thorough analysis of the MRI data, utilizing the CDM Explore platform to compare MRI results with changes in neuroinflammatory biomarkers, qEEG, and cognitive test results.
Mechanism of Action and Preclinical Data
NTRX-07 is a small orally available molecule that targets receptors that increase in neurodegenerative diseases, including Alzheimer's disease, amyotrophic lateral sclerosis, multiple sclerosis, and chronic pain syndromes. In various animal models of Alzheimer's disease, NTRX-07 demonstrated the ability to restore normal function of microglia, the key immune cells in the brain.
This restoration of microglial function led to decreased microglial-induced inflammation, reduced levels of the Alzheimer's-associated Aβ peptide in the brain, and substantially improved neuronal synaptic plasticity, learning, and memory in preclinical studies.
Future Development Plans
The positive results have generated significant interest for future development.
