SGT-610 Mechanism and Potential
SGT-610, a hedgehog signaling pathway (搜索) blocker, has the potential to become the first ever treatment for prevention of BCCs in Gorlin syndrome (搜索) patients, if approved. The drug contains patidegib as its active substance, designed to block the smoothened, frizzle class receptor (SMO (搜索)) signal, allowing cells to function normally and reducing new tumor production.
Understanding Gorlin Syndrome
Gorlin syndrome (搜索) affects approximately 1 in 27,000-31,000 people in the U.S. and represents an autosomal dominant genetic disorder mostly caused by inheritance of one defective copy of the tumor suppressor patched homolog 1 (PTCH1 (搜索)) gene. Under normal conditions, the PTCH1 gene blocks the SMO (搜索) gene, turning off the hedgehog signaling pathway (搜索) when not needed. However, mutations in the PTCH1 gene may cause loss of PTCH1 function, release of SMO, and allow BCC tumor cells to divide uncontrollably.
