The study is being conducted in collaboration with Dr. Antoinette Maassen van den Brink, Professor of Neurovascular Pharmacology at Erasmus University Medical Center, who serves as principal investigator and sponsor. The research will utilize a trigeminal neurovascular reactivity model, measuring forehead skin blood flow responses to capsaicin and electrical stimulation using Laser Speckle Contrast Imaging (LSCI).

TNX-1900 represents a distinct therapeutic approach compared to existing migraine (搜索) treatments. The study employs LSCI, a newer detection method for dermal blood flow that Dr. Maassen van den Brink's team recently validated in the trigeminal neurovascular reactivity model. This technology provides real-time and higher resolution dermal blood flow measurements compared to Laser Doppler Perfusion Imaging used in earlier studies.

Previous research by her team demonstrated that both CGRP inhibitors and triptans can inhibit forehead dermal blood flow responses to capsaicin in migraineurs and healthy volunteers.
Drug Formulation and Safety Profile
TNX-1900 is formulated as a drug-device combination product utilizing an intranasal actuator device for oxytocin delivery. The formulation includes magnesium, which enhances oxytocin receptor (搜索) binding and provides inhibitory effects on trigeminal neurons with resultant craniofacial analgesic effects, as demonstrated in animal models.
When delivered nasally, oxytocin concentrates in the trigeminal system, resulting in targeted binding to receptors on trigeminal neurons. This targeted delivery approach results in low systemic exposure and potentially lower risk of off-target effects compared to systemic CGRP inhibition.
